Just double click it, choose the mastery type you want (tyria, HoT, PoF etc.), and it'll give you that item. It autoconsumes or you double click that one as well.
And yes, select a mastery to train before using it. If you want HoT masteries, select one mastery from HoT, then when you use that item it'll give you XP for that mastery track.
