Overlord RainyDay.2084
I'm guessing by main quest, you mean the personal story? You don't need to finish that before you get your glider. You can freely switch between stories, and it will even remember what chapters you've completed so you can pick back up where you left off. You should just need to finish the first 2 parts of the story (Prologue: Rally to Maguuma, and Torn from the Sky) to get the glider. Do note that you will need to train the first level of the mastery to use it and only XP gained on HoT or LS3 maps will work. All the masteries are account-wide unlocks, so once you have the glider, all your characters can use it.

Overlord RainyDay.2084
On your hero menu, there should be a tab called story journal. You can switch between unlocked story chapters there. I believe you have to be 80 to start HoT, but if you used a booster you should be set.Just a warning, heart of thorns is a SIGNIFICANT jump in difficulty. If you're only used to central tyria, and especially if you haven't done any lvl 80 stuff yet, you may be in for a shock. Pocket Raptors and Mordrem snipers don't play around. FogLeg.9354
Glider and all other masteries are shared between all your characters. To unlock Basic Glider you need to finish first step of HoT story and get access to the Verdant Brink map. Killing enemies in that map (and later other Heart of Maguuma maps) will earn you xp that will slowly fill up your xp bar. Once it is full you can upgrade one of the masteries. But each mastery also requires finishing certain number Hero Points, some of these are just about finding and reaching them but there are several very tough to solo so be ready to ask help in map chat and also look out for others asking for help to do them. Why HoT and its maps are universally most hated from all GW2 content is that the difficulty jump from Central Tyria is huge (yes they have been nerfed already multiple times to somewhat compensate lack of players there). Every enemy in HoT is basically veteran from base game and every veteran in HoT is champ from Tyria. It does not help that they tend to come in large groups. Thing is, all these maps were designed to be played in large groups, there are map-wide meta events to be finished and big bosses to be slayed, and all this requires full map, several commanders doing multiple events same time and in general understanding what you doing. These days only couple maps are farmed at specific times, off hours they are abandoned and you can not really do anything there alone. It does not help that without masteries like Advanced Gliding or Mushroom Jumping you can not even reach most of map, but because it has multiple layers vertically you will never know how some point can be reached unless you follow a specific guide.
