Do Sanctuary Runes provide barrier/healing while in Reaper Shroud?

Yes. It does work.

But in my opinion it's a wasted rune on reaper.

Mainly because barrier doesn't prevent shroud from degeneration

Not directly no but it does in an indirect manner. It covers the damage LF would have otherwise been reduced by.
