To gain masteries you need to own either Heart of Thorns or Path of Fire and do the first part of the expansion story to unlock the mastery system. Some masteries are available to train right away, others need to be unlocked individually - for example the PoF masteries are all linked to mounts and unlock when you obtain the relevant mount.

You need to choose mastery tracks to train in the menu in the Hero Panel. When you're in relevant maps any XP you gain will go towards the mastery you've chosen - HoT masteries can only be trained up in HoT maps, core Tyria ones in base game maps and so on. When it's full you need to spend mastery points to unlock it. Points come from completing storylines and achievements (you can see which ones give masteries because they're marked with coloured stars in the achievement menu).

 

Once a mastery is unlocked it's available to all characters on your account forever, so you only need to train each one once.
